Retatrutide is an investigational synthetic peptide designed to activate three distinct receptor systems within a single molecule. Its pharmacological profile combines activity at the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor. This arrangement places it within a broader class of agents often described as multi-agonists, which contrast with compounds that engage one or two targets. Research interest centers on whether simultaneous signaling produces effects that single-receptor agonists cannot achieve alone. A single molecular entity also simplifies manufacturing and delivery logistics compared with combining separate agents.
Mechanistic proposals link each receptor to a different physiological role. Activation of the glucose-dependent insulinotropic polypeptide and glucagon-like peptide-1 receptors is associated with reduced appetite, slower gastric emptying, and glucose-dependent insulin release. Glucagon receptor signaling, by contrast, is associated with increased energy expenditure and altered lipid handling, though it can also raise blood glucose. The design intent is to balance these contributions so that weight reduction is enhanced without unacceptable glycemic trade-offs. How well that balance holds across individuals is not fully resolved.
Published information places retatrutide in clinical development rather than on the market as an approved therapy. Early-stage and mid-stage trials have examined tolerability and changes in body weight, and larger studies continue to report results over time. Open questions include the durability of effects after treatment stops, the composition of weight lost, and cardiovascular outcomes over long periods. Statements about definitive benefit should therefore be treated as provisional. Regulatory status varies by jurisdiction and changes as applications are reviewed.
The peptide backbone is chemically modified to resist rapid enzymatic breakdown in the body. A fatty acid side chain promotes binding to serum albumin, which slows renal clearance and supports an extended circulation time. These modifications allow less frequent administration than would be possible with an unmodified peptide. The precise contribution of glucagon receptor activation to the overall metabolic effect remains an area of active investigation, because glucagon raises glucose while also increasing energy expenditure.
Development has progressed through early- and mid-stage human studies in adults with obesity and with type 2 diabetes. Published phase 2 data reported reductions in body weight and improvements in glycemic markers over the treatment period. No regulatory agency has approved the compound for any indication, and it remains available only within controlled research settings. Whether benefits observed in trials translate into durable outcomes after treatment stops is not yet established.

Clinical development has advanced through phase 2 trials in adults with obesity and type 2 diabetes. Reported phase 2 results described substantial average weight reduction over roughly forty-eight weeks of weekly dosing. A phase 3 program is ongoing to confirm efficacy and assess long-term safety. Because the compound has not received regulatory approval, it is not available as a prescription product. Public discussion of retatrutide often conflates trial findings with marketed status, an important distinction when interpreting coverage of the topic.
Retatrutide is a synthetic peptide developed as a single molecule that activates three distinct hormone receptors: GLP-1, GIP, and glucagon. The compound carries the internal designation LY3437943 and was engineered by modifying the backbone of glucose-dependent insulinotropic polypeptide. Its sequence incorporates non-natural amino acids and a fatty acid side chain that extends circulation time. The triple-agonist design aims to combine appetite suppression, improved insulin response, and increased energy expenditure in one agent. Published reports describe it as an investigational product rather than an approved medicine.
Each receptor contributes a different physiological effect. Activation of the GLP-1 receptor slows gastric emptying and reduces appetite signaling in the brain. GIP receptor activity influences insulin secretion and lipid handling, while glucagon receptor stimulation raises energy use and fat oxidation. Combining these pathways is intended to produce weight loss beyond what single- or dual-receptor agonists achieve. Researchers attribute the observed potency to simultaneous engagement of all three targets, though the exact contribution of each receptor to overall effect remains under investigation.
Development has progressed from single- and multiple-ascending-dose studies in healthy volunteers into larger randomized trials in adults with obesity, type 2 diabetes, and fatty liver disease. Early reports describe dose-dependent reductions in body weight and improvements in glycemic markers over treatment periods of several months. Whether the glucagon arm adds tolerability cost without added benefit is still debated. Long-term cardiovascular outcomes, effects after treatment stops, and performance in older or comorbid populations are open questions rather than settled findings. Approval status may change, so the current investigational label should be confirmed against regulatory sources.
Retatrutide is an investigational synthetic peptide developed under the code LY3437943, with a backbone derived from glucose-dependent insulinotropic polypeptide. Several non-proteinogenic residues, including alpha-aminoisobutyric acid, appear in that backbone, and a fatty diacid side chain attached through a linker extends circulation time. The molecule carries roughly thirty-nine amino acid units and a total mass near 4.7 kilodaltons. Administration is by subcutaneous injection once weekly. Published work uses both the name retatrutide and the code LY3437943.
Peptides in this class degrade mainly through hydrolysis, oxidation, and aggregation. The lipid modification improves plasma residence time but can also promote self-association in aqueous solution at higher concentrations. Oxidation of methionine and deamidation of asparagine residues are common chemical liabilities that accumulate during storage. Stability studies therefore track purity loss, aggregate formation, and changes in receptor-binding potency over time under defined temperature and humidity conditions.
Solid peptide is generally held as a lyophilised powder at low temperature to slow degradation, with desiccant to limit moisture uptake. Reconstituted solutions are less stable and are usually kept refrigerated and protected from light for short periods. Repeated freeze-thaw cycles are avoided because they encourage aggregation. Laboratory handling includes work in a fume hood or laminar flow cabinet to limit inhalation and contamination. Weighing and transfer steps are performed with antistatic tools to reduce static-driven loss of fine powder.
Characterising a peptide of this size relies on a combination of chromatographic and mass spectrometric techniques. Reversed-phase high-performance liquid chromatography separates the intact molecule from related impurities, while electrospray mass spectrometry confirms molecular mass and detects truncation or oxidation products. Peptide mapping after enzymatic digestion verifies the amino acid sequence and locates modified residues. Because the molecule carries a lipid chain, assays must also distinguish the correctly conjugated product from incompletely acylated species.
Depending on manufacturer, the dressings are available in transparent, translucent, or opaque varieties. Certain brands will develop a wider opaque area when saturated with exudate, which is sometimes a suggested indicator of when to replace them. The moist conditions produced under the dressing are intended to promote wound healing, including fibrinolysis and angiogenesis, without causing softening and breakdown of tissue. Most hydrocolloid dressings are water-resistant, allowing gentle washing and bathing.
==== Physical ==== Heavy, long-term exposure to marijuana may have physical, mental, behavioral and social health consequences. It may be "associated with diseases of the liver (particularly with co-existing hepatitis C), lungs, heart, and vasculature". A 2014 review found that while cannabis use may be less harmful than alcohol use, the recommendation to substitute it for problematic drinking was premature without further study. Various surveys conducted between 2015 and 2019 found that many users of cannabis substitute it for prescription drugs (including opioids), alcohol, and tobacco; most of those who used it in place of alcohol or tobacco either reduced or stopped their intake of the latter substances. Cannabinoid hyperemesis syndrome (CHS) is a severe condition seen in some chronic cannabis users where they have repeated bouts of uncontrollable vomiting for 24–48 hours. Four cases of death have been reported as a result of CHS. A limited number of studies have examined the effects of cannabis smoking on the respiratory system. Chronic heavy marijuana smoking is associated with respiratory infections, coughing, production of sputum, wheezing, and other symptoms of chronic bronchitis. The available evidence does not support a causal relationship between cannabis use and chronic obstructive pulmonary disease. Short-term use of cannabis is associated with bronchodilation. Other side effects of cannabis use include cannabinoid hyperemesis syndrome (CHS), a condition which involves recurrent nausea, cramping abdominal pain, and vomiting.
== Controversy == In 2020, Horsley was involved in a Twitter feud. In 2021, Horsley had a dispute with fellow Hamden councilmember Justin Farmer, when she asked that he address her as "doctor", which was requested by the Hamden Council President a few months earlier in solidarity with Jill Biden. Her resignation was requested but Horsley finished her term. Despite this political opposition, Horsley has actively supported diversity in science as a founding co-Chair of Society of Investigative Dermatology's Diversity Committee.
Moreover, the ability to perform high frequency fractionation immediately from the eluent of a nano-LC column has greatly increases peak resolution and improved the overall separation quality when compared to continuous flow nano-LC devices. An HPLC column was first built directly into a microfluidic device by using TPE instead of PDMS for the device fabrication. The additional strength of TPE made it capable of supporting the higher pressures needed for HPLC such that a single, microfluidic lab-chip could perform chemical separation, fractionation, and further droplet manipulation. In order to increase the quality of chromatographic output, sturdier devices made of glass have shown the ability to withstand far greater pressure than TPE. Achieving these higher pressures to increase the degree of separation and eliminating all dead volumes through immediate droplet formation has shown the potential for droplet microfluidics to expand and improve the capabilities of HPLC separations.

=== Announcement === Prompted by the government, the Rhodesian Broadcasting Corporation told the public to stand by for an important announcement from the Prime Minister at 13:15 local time. Smith went first to Government House to inform Gibbs that his Cabinet had declared independence, then to Pockets Hill Studios in east Salisbury to announce UDI to the nation. He read the proclamation aloud, then stated that independence had been declared because it had become "abundantly clear that it is the policy of the British government to play us along with no real intention of arriving at a solution which we could possibly accept ... I promised the people of this country that I would continue to negotiate to the bitter end and that I would leave no stone unturned in my endeavours to secure an honourable and mutually accepted settlement; it now falls to me to tell you that negotiations have come to an end". Smith said that he believed that he would be remiss in his duty if he allowed Rhodesia to continue to "drift in its present paralysing state of uncertainty", and that following Britain's abandonment of the Federation his government was determined that "the same will never be allowed to happen here". He claimed that UDI did not mark "a diminution in the opportunities which our African people have to advance and prosper in Rhodesia", described "racial harmony in Africa" as part of his agenda and condemned black Rhodesian activities as attempts to "blackmail the British government into ... handing the country over to irresponsible rule".
Because lichens accumulate pollutants over time, they allow for high-density spatial sampling, even in areas lacking automated monitoring stations. This makes X. parietina particularly useful for retrospective pollution assessments and long-term environmental monitoring. Findings from the Adriatic Italy study support its role as a reliable tool for tracking pollution trends, identifying pollutant sources, and informing environmental planning and public health strategies. Environmental influences on growth rate have important implications for biomonitoring studies that use X. parietina to assess air quality. Because the lichen accumulates pollutants over time, differences in growth rates between populations can affect pollutant load measurements. Thalli in humid, cooler environments may accumulate contaminants at a lower rate than those in drier, warmer habitats simply due to differences in growth and dilution effects. Consequently, lichen-based biomonitoring efforts must consider local climatic conditions to ensure accurate comparisons of atmospheric pollution levels across regions.
==== Canada ==== Health Canada has investigated the risks and benefits of pseudoephedrine and ephedrine/Ephedra. Near the end of the study, Health Canada issued a warning on their website stating that those who are under the age of 12, or who have heart disease and may have strokes, should avoid taking pseudoephedrine and ephedrine. Also, they warned that everyone should avoid taking ephedrine or pseudoephedrine with other stimulants like caffeine. They also banned all products that contain both ephedrine (or pseudoephedrine) and caffeine. Products whose only medicinal ingredient is pseudoephedrine must be kept behind the pharmacy counter. Products containing pseudoephedrine along with other medicinal ingredients may be displayed on store shelves but may be sold only in a pharmacy when a pharmacist is present. Pseudoephedrine is a Schedule VI Class A precursor in Canada under the Controlled Drugs and Substances Act.
These names were later suggested for element 114 (flerovium) and element 116 (moscovium). Flerovium became the name of element 114; the final name proposed for element 116 was instead livermorium, with moscovium later being proposed and accepted for element 115 instead. Traditionally, the names of all noble gases end in "-on", with the exception of helium, which was not known to be a noble gas when discovered. The IUPAC guidelines valid at the moment of the discovery approval however required all new elements be named with the ending "-ium", even if they turned out to be halogens (traditionally ending in "-ine") or noble gases (traditionally ending in "-on"). While the provisional name ununoctium followed this convention, a new IUPAC recommendation published in 2016 recommended using the "-on" ending for new group 18 elements, regardless of whether they turn out to have the chemical properties of a noble gas. The scientists involved in the discovery of element 118, as well as those of 117 and 115, held a conference call on 23 March 2016 to decide their names. Element 118 was the last to be decided upon; after Oganessian was asked to leave the call, the remaining scientists unanimously decided to have the element "oganesson" after him. Oganessian was a pioneer in superheavy element research for sixty years reaching back to the field's foundation: his team and his proposed techniques had led directly to the synthesis of elements 107 through 118.
=== Gender dysphoria === Many international guidelines and institutions require persistent, well-documented gender dysphoria as a pre-requisite to starting gender-affirmation therapy. Gender dysphoria refers to the psychological discomfort or distress that an individual can experience if their sex assigned at birth is incongruent with that person's gender identity. Signs of gender dysphoria can include comorbid mental health stressors such as depression, anxiety, low self-esteem, and social isolation. Not all gender nonconforming individuals experience gender dysphoria, and measuring a person's gender dysphoria is critical when considering medical intervention for gender nonconformity.

It does not form a hydrate, unlike sodium sulfate. The salt crystallizes as double six-sided pyramids, classified as rhombic. They are transparent, very hard and have a bitter, salty taste. The salt is soluble in water, but insoluble in solutions of potassium hydroxide (sp. gr. 1.35), or in absolute ethanol.
=== Expansion === The idea of expanding the tournament had been suggested as early as 2013 by then-UEFA president Michel Platini, as well as in 2016 by current FIFA president Gianni Infantino. Opponents of the proposal argued that the number of matches played was already at an unacceptable level, that the expansion would dilute the quality of the matches, and that the decision was driven by political rather than sporting concerns, specifically that Infantino was trying to win his election by promising to bring more countries to the World Cup. Starting with the 2026 edition, the World Cup expanded to 48 teams, an increase of 16 teams compared to the previous seven tournaments. As approved by the FIFA Council on March 14, 2023, the teams were split into 12 groups of 4 teams, with the top 2 teams in each group and the 8 best third-place teams progressing to a new round of 32. It was the first expansion and format change since 1998. The total number of matches played increased from 64 to 104, and the number of matches played by teams reaching the semifinals increased from 7 to 8. The tournament lasted 39 days, an increase from the 32 days of the 2014 and 2018 tournaments, and from the 29 days of the 2022 tournament, although each team still played three group matches. The final matchday at club level for players named in the final squads was May 24, 2026; clubs had to release their players by May 25, with exceptions granted to players participating in continental club competition finals up until May 30.
== Chemical properties == Following is a table listing the one-letter symbols, the three-letter symbols, and the chemical properties of the side chains of the standard amino acids. The masses listed are based on weighted averages of the elemental isotopes at their natural abundances. Forming a peptide bond results in elimination of a molecule of water. Therefore, the protein's mass is equal to the mass of amino acids the protein is composed of minus 18.01524 Da per peptide bond.
Overfishing of fish species targeted for their fish bladders has led to fish stock collapse. The first major population decline from the swim bladder trade was of the Chinese bahaba or giant yellow croaker Bahaba taipingensis of the China Seas. While Chinese bahaba fishing was banned for domestic trade in 1989 after the classification of the fish as Class II under the Wildlife Protection Law of the People's Republic of China, the Chinese bahaba population continued to decline and was added to the IUCN Red List in 2009. The totoaba began to be targeted for its perceived similarity to the Chinese bahaba by the 1920s, and increased in scope until it was categorized as a vulnerable species. On Lake Victoria, there is increased usage of illegal monofilament nets and targeting of undersized fish, impacting the Nile perch population. Kenya, Uganda and Tanzania have beach patrols and fishing regulations to regulate the fish maw trade.

It is described as a single molecule that acts at three receptors: the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor. This triple activity distinguishes it from agents that target one or two of these pathways.
It is characterized in the literature as an investigational agent under clinical study. Approval status depends on jurisdiction, and readers should check current regulatory information rather than assume availability.
The combination is intended to pair appetite-related and glucose-related effects with mechanisms that increase energy expenditure. Whether the combined profile offers advantages over simpler agonists is the subject of ongoing research.
