== Prognosis == The use of DMARDs in the management of JIA have made disease-free remission achievable for the majority of children with JIA. === Lactic acid hypothesis === It was once believed that lactic acid build-up was the cause of muscle fatigue. The assumption was lactic acid had a "pickling" effect on muscles, inhibiting their ability to contract. The impact of lactic acid on performance is now uncertain, it may assist or hinder muscle fatigue. Produced as a by-product of fermentation, lactic acid can increase intracellular acidity of muscles. This can lower the sensitivity of contractile apparatus to calcium ions (Ca2+) but also has the effect of increasing cytoplasmic Ca2+ concentration through an inhibition of the chemical pump that actively transports calcium out of the cell. This counters inhibiting effects of potassium ions (K+) on muscular action potentials. Lactic acid also has a negating effect on the chloride ions in the muscles, reducing their inhibition of contraction and leaving K+ as the only restricting influence on muscle contractions, though the effects of potassium are much less than if there were no lactic acid to remove the chloride ions. Ultimately, it is uncertain whether lactic acid reduces fatigue through increased intracellular calcium or increases fatigue through reduced sensitivity of contractile proteins to Ca2+.

=== Isotope labeling measuring techniques === Any technique in measuring the difference between isotopomers can be used. The two primary methods, nuclear magnetic resonance (NMR) and mass spectrometry (MS), have been developed for measuring mass isotopomers in stable isotope labeling. Proton NMR was the first technique used for 13C-labeling experiments. Using this method, each single protonated carbon position inside a particular metabolite pool can be observed separately from the other positions. This allows the percentage of isotopomers labeled at that specific position to be known. The limit to proton NMR is that if there are n carbon atoms in a metabolite, there can only be at most n different positional enrichment values, which is only a small fraction of the total isotopomer information. Although the use of proton NMR labeling is limiting, pure proton NMR experiments are much easier to evaluate than experiments with more isotopomer information. In addition to Proton NMR, using 13C NMR techniques will allow a more detailed view of the distribution of the isotopomers. A labeled carbon atom will produce different hyperfine splitting signals depending on the labeling state of its direct neighbors in the molecule. A singlet peak emerges if the neighboring carbon atoms are not labeled. A doublet peak emerges if only one neighboring carbon atom is labeled. The size of the doublet split depends on the functional group of the neighboring carbon atom.
=== Identifying Translated mRNA Regions === By using specific drugs, ribosome profiling can identify initiating regions of mRNA, elongating regions, and areas of translation stalling. Initiating regions can be detected by adding harringtonine or lactidomycin to prevent any further initiation. This allows the starting codon of the mRNAs throughout the cell lysate to be analyzed, which has been used to determine non-AUG sequences that do initiate translation. The other elongating regions can be detected by adding antibiotics like cycloheximide that inhibit translocation, chloramphenicol that inhibits transfer of peptides within the ribosome, or non-drug means like thermal freezing. These elongation freezing methods allow for the kinetics of translation to be analyzed. Since multiple ribosomes can translate a single mRNA molecule to speed up the translation process, RiboSeq demonstrates the protein coding regions within the mRNA and how quickly this is done depending on the mRNA being sequenced. This also allows for ribosome profiling to show pause sites within the transcriptome at specific codons. These sites of slow or paused translation are demonstrated by an increase in ribosome density and these pauses can link specific proteins with their roles within the cell.
=== Irritation === Irritation and itching of the vulva is called pruritus vulvae. This can be a symptom of many disorders, some of which may be determined by a patch test. The most common cause of irritation is thrush, a fungal infection. Vulvovaginal health measures can help to prevent many disorders including thrush. Infections of the vagina such as vaginosis and of the uterus may produce vaginal discharge, which can be an irritant when it comes into contact with the vulvar tissue. Inflammation as vaginitis, vulvovaginitis and vulvitis can result from this causing irritation and pain. Ingrown hairs resulting from pubic hair shaving can cause folliculitis where the hair follicle becomes infected; or give rise to an inflammatory response known as pseudofolliculitis pubis. A less common cause of irritation is genital lichen planus, another inflammatory disorder. A severe variant of this is vulvovaginal gingival syndrome, which can lead to narrowing of the vagina, or vulva destruction. Many types of infection and other diseases including some cancers may cause irritation.
== Human rights abuses == Throughout the transitional period, the Transitional Government of Ethiopia was criticized by various human rights organizations for abuses ranging from extrajudicial executions to unlawful detentions. At the establishment of the TGE in 1991, when the EPRDF first took power, human rights organizations such as Human Rights Watch and Amnesty International expressed optimism about the future of the state of human rights in Ethiopia. During the previous regime under Mengistu, human rights groups could not exist; following May 1991, however, human rights watchdogs such as the Ethiopian Human Rights Council, the Ethiopian Congress for Democrats, and the human rights committee of the Committee of Eleven were established. However, hopes were quickly dashed following a pattern of rights violations aimed at political dissidents across the country. For instance, at least ten demonstrators in Addis Ababa were killed while protesting the EPRDF in their early days of power. In addition, an estimated 5,000-100,000+ members (including jailed former soldiers) of the previous PDRE were swiftly imprisoned under the TGE. While some were later released, many others were held without being officially charged or having a trial. Members of Mengistu's Workers' Party of Ethiopia were also not permitted to travel abroad or go back to work if they were previously detained by the EPRDF for the duration of the year, but generally found themselves able to do so in 1992.
== Concept of disease == In general, disease is perceived as a disharmony (or imbalance) in the functions or interactions of yin, yang, qi, xuĕ, zàng-fǔ, meridians etc. and/or of the interaction between the human body and the environment. Therapy is based on which "pattern of disharmony" can be identified. Thus, "pattern discrimination" is the most important step in TCM diagnosis. It is also known to be the most difficult aspect of practicing TCM. To determine which pattern is at hand, practitioners will examine things like the color and shape of the tongue, the relative strength of pulse-points, the smell of the breath, the quality of breathing or the sound of the voice. For example, depending on tongue and pulse conditions, a TCM practitioner might diagnose bleeding from the mouth and nose as: "Liver fire rushes upwards and scorches the Lung, injuring the blood vessels and giving rise to reckless pouring of blood from the mouth and nose." He might then go on to prescribe treatments designed to clear heat or supplement the Lung.
