Clinical development has progressed through phase 2 trials in adults with obesity and type 2 diabetes, with phase 3 programs reported as ongoing. Reported outcomes include reductions in body weight and improvements in glycemic measures over defined treatment periods. Whether these effects translate into durable benefits after treatment ends remains an open question. Long-term safety data across broad populations are not yet complete, and regulatory decisions have not been announced.
Retatrutide is an investigational synthetic peptide engineered to activate three distinct hormone receptors within a single molecule. It targets the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor simultaneously. This triagonist design distinguishes it from earlier incretin-based compounds that act on one or two of these pathways. Structural modifications relative to native gut hormones extend its residence time in circulation. The molecule remains under clinical evaluation and is not approved for any indication.
Receptor activation produces downstream effects that differ by tissue. GLP-1 receptor signaling influences appetite regulation and insulin secretion in a glucose-dependent manner. GIP receptor activity contributes to metabolic handling of nutrients and may modulate adipose tissue. Glucagon receptor engagement raises energy expenditure and promotes hepatic lipid turnover, though the balance among these actions in humans is still being characterized. Preclinical models showed reductions in body weight and improved glycemic markers.

Several questions about the compound remain unresolved. The durability of weight reduction after treatment stops, the frequency of gastrointestinal side effects, and the long-term cardiovascular profile are topics of ongoing study. Regulatory submissions and phase 3 trial outcomes have not been fully reported in the public literature. Because most available data come from controlled trials rather than general-population use, conclusions about effectiveness outside study settings are provisional. The distinction between established findings and open questions matters when interpreting early coverage of the drug.
Retatrutide is an investigational peptide developed by a pharmaceutical company as a multi-receptor agonist for treating obesity and type 2 diabetes. The compound emerged from research into gut-hormone analogues that act on several receptors simultaneously rather than on a single target. Early preclinical work examined how combined activity at three distinct receptors might produce greater metabolic effects than single-receptor compounds. Published phase 2 results have described substantial reductions in body weight among participants, although the compound remains unapproved in most jurisdictions as of the mid-2020s.
Peptides in this class degrade mainly through hydrolysis, oxidation, and aggregation. The lipid modification improves plasma residence time but can also promote self-association in aqueous solution at higher concentrations. Oxidation of methionine and deamidation of asparagine residues are common chemical liabilities that accumulate during storage. Stability studies therefore track purity loss, aggregate formation, and changes in receptor-binding potency over time under defined temperature and humidity conditions.
Solid peptide is generally held as a lyophilised powder at low temperature to slow degradation, with desiccant to limit moisture uptake. Reconstituted solutions are less stable and are usually kept refrigerated and protected from light for short periods. Repeated freeze-thaw cycles are avoided because they encourage aggregation. Laboratory handling includes work in a fume hood or laminar flow cabinet to limit inhalation and contamination. Weighing and transfer steps are performed with antistatic tools to reduce static-driven loss of fine powder.
A refrigerator truck or chiller lorry (also called a reefer), is a van or truck designed to carry perishable freight at low temperatures. Most long-distance refrigerated transport by truck is done in articulated trucks pulling refrigerated hardside (box) semi-trailers, although insulated curtainsiders are common in some countries. Occasionally, refrigerated trailers have been used as temporary morgues, and second-hand refrigerated trailers are frequently sold for use in tiny home conversions due to their insulation and existing status as a vehicle.
India is a federal union comprising 28 states and 8 union territories. All states, as well as the union territories of Jammu and Kashmir, Puducherry and the National Capital Territory of Delhi, have elected legislatures and governments following the Westminster system. The remaining five union territories are directly ruled by the central government through appointed administrators. In 1956, under the States Reorganisation Act, states were reorganised on a linguistic basis. Over a quarter of a million local government bodies operate at the city, town, district, block, and village levels.
Minimally invasive surgery (also known as minimally invasive procedure) encompass surgical techniques that limit the size of incisions needed, thereby reducing wound healing time, associated pain, and risk of infection. Surgery by definition is invasive, and many operations requiring incisions of some size are referred to as open surgery. Incisions made during open surgery can sometimes leave large wounds that may be painful and take a long time to heal. Advancements in medical technologies have enabled the development and regular use of minimally invasive procedures. For example, endovascular aneurysm repair, a minimally invasive surgery, has become the most common method of repairing abdominal aortic aneurysms in the US as of 2003. The procedure involves much smaller incisions than the corresponding open surgery procedure of open aortic surgery. Interventional radiologists were the forerunners of minimally invasive procedures. Using imaging techniques, radiologists were able to direct interventional instruments through the body by way of catheters instead of the large incisions needed in traditional surgery. As a result, many conditions once requiring surgery can now be treated non-surgically. Diagnostic techniques that do not involve incisions, puncturing the skin, or the introduction of foreign objects or materials into the body are known as non-invasive procedures. Several treatment procedures are classified as non-invasive. A major example of a non-invasive alternative treatment to surgery is radiation therapy, also called radiotherapy.
In June 1963, a new terminal built for the jet age was dedicated, and the airport was rechristened Memphis Metropolitan Airport. United Nations ambassador Adlai Stevenson II spoke at the inauguration ceremony and replaced the ribbon cutting with a rocket launch. The terminal was designed by the local firm Mann & Harrover and became Roy Harrover's most famous work. It was one of the first airport terminals to have a two-level design where passengers boarded aircraft via jet bridges on the upper level and collected their luggage on the lower level. Another unique aspect of the structure was its columns resembling martini glasses. It opened to passengers in July and was part of an expansion project that also included a control tower and a north–south runway. In 1969, the Memphis–Shelby County Airport Authority was formed, and the facility changed its name to Memphis International Airport after being designated a customs port of origin. Expansion continued in the 1970s. A west concourse was added in 1974 and an east one the following year. The airport constructed another north–south runway, a parking garage, and a road linking the terminal to Interstate 240. The central concourse was extended as well. In 1988, the latter concourse was named B, the west one A, and the east one C.

After the blood is collected, the bottles are incubated at body temperature to encourage the growth of microorganisms. Bottles are usually incubated for up to five days in automated systems, although most common bloodstream pathogens are detected within 48 hours. The incubation time may be extended further if manual blood culture methods are used or if slower-growing organisms, such as certain bacteria that cause endocarditis, are suspected. In manual systems, the bottles are visually examined for indicators of microbial growth, which might include cloudiness, the production of gas, the presence of visible microbial colonies, or a change in colour from the digestion of blood, which is called hemolysis. Some manual blood culture systems indicate growth using a compartment that fills with fluid when gases are produced, or a miniature agar plate which is periodically inoculated by tipping the bottle. To ensure that positive blood cultures are not missed, a sample from the bottle is often inoculated onto an agar plate (subcultured) at the end of the incubation period regardless of whether or not indicators of growth are observed. In developed countries, manual culture methods have largely been replaced by automated systems that provide continuous computerized monitoring of the culture bottles. These systems, such as the BACTEC, BacT/ALERT and VersaTrek, consist of an incubator in which the culture bottles are continuously mixed.
A biosynthetic pathway has been hypothesized based on proposed gene functions (see figure). btmM, with homology to Zn+2 aminopeptidases, is predicted to cleave the N-terminal methionine residue, which is not present in the bottromycin final product. btmE and btmF both contain YcaO-like domains. It is believed that one Although it is unclear which enzyme is responsible for which step, it is hypothesized that one catalyzes macrocyclic amidine formation while the other catalyzes thiazoline formation. btmJ, encoding an enzyme with cytochrome P450 homology, may oxidize the thiazoline to the thiazole. btmH or btmI both have homology to hydrolytic enzymes (α/β hydrolase and metallo-dependent hydrolase, respectively) may catalyze follower peptide hydrolysis. An alternative proposed role for btmH or btmI is to function as a cyclodehydratase in macrocyclization. Gene deletion studies failed to elucidate the function of other proteins within the cluster.
== Medical uses == Sertraline has been approved for major depressive disorder, obsessive–compulsive disorder (OCD), post-traumatic stress disorder (PTSD), premenstrual dysphoric disorder, panic disorder, social anxiety disorder (SAD), and generalized anxiety disorder (GAD). In August 2008, Obama and Biden met in secret to discuss the possibility of a place for Biden in the Obama administration, and developed a strong personal rapport. On August 22, Obama announced that Biden would be his running mate. The New York Times reported that the choice reflected a desire for someone with foreign policy and national security experience. Others pointed out Biden's appeal to middle-class and blue-collar voters. Biden was officially nominated for vice president on August 27 at the 2008 Democratic National Convention in Denver. Biden's vice-presidential campaigning gained little media attention, as the press devoted far more coverage to the Republican nominee and then-governor of Alaska, Sarah Palin. Under instructions from the campaign, Biden kept his speeches succinct and tried to avoid offhand remarks. Privately, Biden's remarks frustrated Obama. "How many times is Biden gonna say something stupid?", he once angrily asked. Obama campaign staffers called Biden's blunders "Joe bombs" and kept Biden uninformed about strategy discussions, which irked Biden. Relations between the two campaigns became strained for a month, until Biden apologized to Obama and the two built a stronger partnership. As the 2008 financial crisis reached a peak in September 2008, and the proposed Emergency Economic Stabilization Act of 2008 became a major factor in the campaign, Biden voted for the $700 billion Emergency Economic Stabilization Act of 2008, which passed in the Senate.
=== Effects on ozone layer === Sulfur dioxide aerosols in the stratosphere can contribute to ozone depletion in the presence of chlorofluorocarbons and other halogenated ozone-depleting substances. The effects of volcanic eruptions containing sulfur dioxide aerosols on the ozone layer are complex, however. In the absence of anthropogenic or biogenic halogenated compounds in the lower stratosphere, depletion of dinitrogen pentoxide in the middle stratosphere associated with its reactivity to the aerosols can promote ozone formation. Injection of sulfur dioxide and large amounts of water vapor into the stratosphere following the 2022 eruption of Hunga Tonga-Hunga Haʻapai resulted in altered atmospheric circulation that promoted a decrease in ozone in the southern latitudes but an increase in the tropics. The additional presence of hydrochloric acid in eruptions can result in net ozone depletion.

In 1942, anatomist Edward Congdon also demonstrated that the right and left mesocolons persisted into adulthood and remained separate from the retroperitoneum—extraretroperitoneal. Radiologist Wylie J. Dodds described this concept in 1986. Dodds extrapolated that unless the mesocolon remained an extraretroperitoneal structure—separate from the retroperitoneum—only then would the radiologic appearance of the mesentery and peritoneal folds be reconciled with actual anatomy. Descriptions of the mesocolon by Toldt, Congdon, and Dodds have largely been ignored in mainstream literature until recently. A formal appraisal of the mesenteric organ anatomy was conducted in 2012; it echoed the findings of Toldt, Congdon, and Dodds. The single greatest advance in this regard was the identification of the mesenteric organ as being contiguous, as it spans the gastrointestinal tract from duodenojejunal flexure to mesorectal level. In 2012 it was discovered that the mesentery was a single organ, which precipitated advancement in colon and rectum surgery and in sciences related to anatomy and development.
N-acetyl Aspartate (NAA): with its major resonance peak at 2.02 ppm, decrease in levels of NAA indicate loss or damage to neuronal tissue, which results from many types of insults to the brain. Its presence in normal conditions indicates neuronal and axonal integrity. Choline: with its major peak at 3.2 ppm, choline is known to be associated with membrane turnover, or increase in cell division. Increased choline indicates increase in cell production or membrane breakdown, which can suggest demyelination or presence of malignant tumors. Creatine and phosphocreatine: with its major peak at 3.0 ppm, creatine marks metabolism of brain energy. Gradual loss of creatine in conjunction with other major metabolites indicates tissue death or major cell death resulting from disease, injury or lack of blood supply. Increase in creatine concentration could be a response to cranialcerebral trauma. Absence of creatine may be indicative of a rare congenital disease. Lipids: with their major aliphatic peaks located in the 0.9–1.5 ppm range, increase in lipids is seen is also indicative of necrosis. These spectra are easily contaminated, as lipids are not only present in the brain, but also in other biological tissue such as the fat in the scalp and area between the scalp and skull. Lactate: Is an AX3 system which results in a doublet (two symmetric peaks) centered about 1.31 ppm, and a quartet (four peaks with relative peak heights of 1:2:2:1) centered about 4.10 ppm.
By the time Smith and Dupont arrived at Government House to see Gibbs, Whitehall had instructed the Governor to formally dismiss Smith and his ministers for treason. Gibbs complied without hesitation. Smith and his ministers ignored this, holding that under the new 1965 constitution Gibbs "no longer ha[d] any executive powers in Rhodesia", and his reserve power to sack them no longer existed. The Rhodesian government hoped that Gibbs might obligingly resign in light of his impotent situation, but he did not; following orders from London, he remained at his post at Government House. Gibbs told the Rhodesian military's senior officers, some of whom were troubled by the perceived choice between Queen and country, to remain at their posts to maintain law and order. He also refused some Army officers' requests for a warrant to arrest Smith. Wilson briefly flirted with the idea of sending Lord Mountbatten to Rhodesia to support Gibbs as a direct representative of the Queen, but this was dropped after Gibbs asked for somebody "higher up" in the royal family instead. "Not likely", Wilson retorted. Prince Philip was suggested by diplomats to be appointed as Governor-General so he could legally sack Smith but this was refused due to the personal risk he would have been in. The Rhodesian government accompanied UDI with emergency measures that it said were intended to prevent alarm, unrest and the flight of people and capital. Press censorship and petrol rationing were imposed, import licences were cancelled and emigration allowances were cut to £100.
== Overdose == In the event of arsenic poisoning (manifesting as seizures, muscle weakness, confusion), the administration of the drug should be immediately discontinued, and appropriate treatment should be initiated. Penicillamine is commonly used at a dose of up to 1 g/day. For patients unable to take oral medications, dimercaprol can be administered intramuscularly at a dose of 3 mg/kg body weight every 4 hours until life-threatening symptoms subside. In cases of coagulopathy, DMSA is recommended at a dose of 10 mg/kg body weight every 8 hours for 5 days, followed by every 12 hours for 2 weeks. Kidney dialysis may also be considered.

It is a synthetic peptide triagonist that engages GIP, GLP-1, and glucagon receptors. Investigational compounds in this class are studied for metabolic conditions rather than for a single organ system.
No regulatory agency has approved it for any indication. It remains an investigational product studied within controlled clinical trial programs.
A dual agonist engages two receptor targets, typically GIP and GLP-1. A triagonist adds glucagon receptor activity, which is intended to influence energy expenditure alongside appetite and glucose handling.
常用质谱测定分子量,再结合肽图或序列分析验证一级结构。单一检测手段一般难以排除结构相近的类似物。多种方法相互印证更为可靠。
